Sequence breakpoints in the aXatoxin biosynthesis gene cluster and Xanking regions in nonaXatoxigenic Aspergillus Xavus isolates

چکیده
Aspergillus Xavus populations are genetically diverse. Isolates that produce either, neither, or both aXatoxins and cyclopiazonic acid (CPA) are present in the Weld. We investigated defects in the aXatoxin gene cluster in 38 nonaXatoxigenic A. Xavus isolates collected from southern United States. PCR assays using aXatoxin-gene-speciWc primers grouped these isolates into eight (A–H) deletion patterns. Patterns C, E, G, and H, which contain 740 kb deletions, were examined for their sequence breakpoints. Pattern C has one breakpoint in the cypA 3 untranslated region (UTR) and another in the verA coding region. Pattern E has a breakpoint in the amdA coding region and another in the ver1 5 UTR. Pattern G contains a deletion identical to the one found in pattern C and has another deletion that extends from the cypA coding region to one end of the chromosome as suggested by the presence of telomeric sequence repeats, CCCTAATGTTGA. Pattern H has a deletion of the entire aXatoxin gene cluster from the hexA coding region in the sugar utilization gene cluster to the telomeric region. Thus, deletions in the aXatoxin gene cluster among A. Xavus isolates are not rare, and the patterns appear to be diverse. Genetic drift may be a driving force that is responsible for the loss of the entire aXatoxin gene cluster in nonaXatoxigenic A. Xavus isolates when aXatoxins have lost their adaptive value in nature. Gene deletion patterns in non-aflatoxigenic isolates of Aspergillus flavus
Fifteen non-aflatoxigenic strains of Aspergillus flavus, represent a wide range of geographic regions of Iran (six provinces include Fars, Ardebil, Guilan, Golestan, Kerman and Semnan) and vegetative compatibility groups (VCGs), were collected from corn (Zea mays L.), peanut (Arachis hypogaea L.) and pistachio (Pistachia vera L.) soils and kernels, were screened for the presence of aflatoxin bi...
متن کاملExpression of aflR, veA and laeA as regulators of aflatoxins and cyclopiazonic acid biosynthesis pathway in Aspergillus flavus
In this study, the production of aflatoxin B1 (AFB1) and cyclopiazonic acid (CPA) was investigated in toxigenic and non-toxigenic Aspergillus flavus with respect to expression of aflR, veA and laeA genes that are involved to toxins production. A. flavus strains were cultured in YES broth at 28 °C for 4 days and the presence of (AFB1) and (CPA) was confirmed and measured by TLC and HPLC. The exp...

To investigate the prevalence of fungal agents in local Egyptian broiler breeder chicken’s premises, tracheal and cloacal swabs from chickens, feed, and water samples were collected. The targeted breed's dead-in-shell eggs and newly hatched chick’s samples were also tested. All fungal isolates were morphologically typed and the predominant fungal species were further subjected to molecular typi...
متن کاملCorrelation of Minimum Inhibitory Concentration Breakpoints and Methicillin Resistance Gene Carriage in Clinical Isolates of Staphylococcus epidermidis
Staphylococcus epidermidis is the most important member of coagulase negative staphylococci responsible for community and hospital acquired infections. Most clinical isolates of S. epidermidis are resistant to methicillin making these infections difficult to treat. In this study, correlation of methicillin resistance phenotype was compared with methicillin resistance (mecA) gene carriage in 55 ...

Pasteurella multocida is known as an important heterogenic bacterial agent causes some severe diseases such as fowl cholera in poultry and haemorrhagic septicaemia in cattle and buffalo. A polymerase chain reaction (PCR) assay was developed using primers derived from conserved part of 16S-23S rRNA gene. The PCR amplified a fragment size of 0.7 kb using DNA from nine avian P. multocida isolates...
